Objective: CLARITY is a new method developed by the Deisseroth group at Stanford (Chung et al., Nature Methods 10: 508-513, 2013). CLARITY (Clear, Lipid-exchanged, Anatomically Rigid, Imaging / Immunostaining compatible, Tissue hYdrogel) is a technique that makes tissues transparent by combining developed neuroscience and chemical engineering.
It is possible to see clearly the long projections of the cells and the subsellular structures in the whole tissue, especially the cells marked with the 3-dimensional display of the transparent tissue, unlike the traditional histological methods, which are done by taking the tissue from the tissue section.
This groundbreaking method for tissue-level imaging has been widely used in many research areas and provides obtaining new findings.
In our course, we aim with this technique to transparentize the mouse spinal cord by passive transparency using a special perfusion and transparency method followed by fluorescent dye staining and 3D imaging under a confocal microscope.
